I have Multi-WAN connection. The second is backup uplink . Does OpenVPN still work if my first WAN (main uplink) fails and the traffic moves to backup uplink?

If you configure your VPN clients to use the second IP as failover, yes.
Tested by me.

On OpenVPN Endian clients add the 2nd IP to the Fallback VPN servers section.
For roaming VPN clients,  add a 2nd IP remote
